SQL Server does not exists or access denided

hi guys ,
i want to coonnect to a sql server on an other machine in a LAN network.
i use of this connection string : "data source=TargetMachine;initial
catalog=mydatabase;password=1;persist security info=True;user id=a;packet
size=4096"
but when i trying to get some data, i get this error : "SQL Server doesn't
exists or access denided"
but i can connect to this database and get data with "Enterprise Manager" or
"Query Analyzer".
what's the problem ?

Most likely, the SQL Server is not set up with mixed mode and you are
connecting with the SQL tools using Windows authentication. This is the most
common reason people have problems, esp. with MSDE.

It could also be the client library being used by the web server.

Or, it could be user based (long shot with a connection string using SQL
security). The easiest way to check the second is try to get data using that
connection string in a Windows App. If it works, then try turning off anon
access in your web app and see if you can get in.
