Define a filter based on record value

I have a datagrid with two columns visible, both being dropdown boxes linked
to other tables.

Is it possible to filter the datasource of the second column, based on the
value chosen in the first column?

Yes. You can set datasource of the second column besed on value chosen in
the first column in datagrid_ItemDataBound event.
