Standard method for slowness investigation

Dear all,

Our company website has got some complaints from our users that they
are unable to login to our website these days.
+ Scenarios:
We have about 25,000 users nationwide in Vietnam to access to our
website via internet with various bandwidth of internet connection line
(ADSL, Leased Line...), even slow type of 64Kbs.
The complaints are that it is very slow to come and login to our site,
these complaints mostly come from local provinces and at month-end.
Note that this only happened started from last month-Febuary.

+ System architecture:
Webserver: MS Windows 2003 Server SP2, IIS
DB Server: SQL Server 2005 Standard Edition
Web server and DB server is hosted inside our company.
Website technology: ASP.NET 2.0

There are a lot of causes to investigate:
- Internet connection at client PC
- internet line to come to our office
- web server or DB server is overload after time
- web application did not optimize for high workload after time

Would you please advise me the standard method of investigation for
this typical type of slowness.

This is a classic asp group.
Dotnet questions should be asked in
<microsoft.public.dotnet.framework.aspnet>

Perhaps this is an IIS problem, which has it's own NG's
